Noradrenaline depleting and blood pressure lowering activity of threo-corbadrine

Abstract Threo-corbadrine caused a long-lasting depletion of noradrenaline in the heart and in mesenteric vessels and lowered the blood pressure of normal and renal hypertensive rats. It is suggested that threo-cobadrine decreases vascular tone by acting peripherally as a substitute adrenergic transmitter.

The action of intravenous injections of pentobarbital and of thiopental on the blood pressure and on the vascular tone in the cortex of the kidney has been studied in albino mice. Pentobarbital caused a pronounced decrease of the filling of blood vessels in the cortex of kidneys together with a long-lasting anaesthesia and a long lasting fall of blood pressure. Thiopental, on the other hand, was less effective on the filling of blood vessels, while its narcotic effect was small and the fall of blood pressure less pronounced. The results indicate that both substances increased the vascular tone in the cortex of the kidney.

4-Hydroxynonenal-Induced Relaxation of Human Mesenteric Arteries1

The effect of 4-hydroxynonenal (4-HNE), a circulating lipid peroxidation product, on the vascular tone of human mesenteric arteries is studied. 4-HNE promotes relaxation of human mesenteric arterial rings in a concentration-dependent manner. Removal of the endothelium or treatment with N(G)-nitro-L-arginine methyl ester hydrochloride (L-NAME; 10(-4) M) partially prevented 4-HNE-induced relaxation, thus suggesting the intervention of nitric oxide from endothelial origin in the vascular effects of 4-HNE.
